UPLC测定黄连中盐酸小檗碱和盐酸巴马汀的含量

摘 要:建立UPLC测定黄连中盐酸小檗碱和盐酸巴马汀含量的方法。采用ACQUITY UPLC BEH-C18色谱柱(2.1 mm×50 mm×1.7 μm),以乙腈-0.05 mol/L磷酸二氢钾溶液为流动相,梯度洗脱,流速为0.2 mL/min,检测波长为350 nm。结果表明盐酸小檗碱和盐酸巴马汀分别在0.037 2~0.297 6 μg/mL,0.008 2~0.065 6 μg/mL的浓度范围内均有良好的线性关系(r=0.999 9),回收率均大于95%,RSD均小于2%。该方法快速、准确、重现性好,可用于测定黄连中盐酸小檗碱和盐酸巴马汀的含量。

UPLC simultaneous determination of berberine hydrochloride and
palmatine hydrochloride in Coptis

Abstract: A method was established for simultaneously determining berberine hydrochloride and palmatine hydrochloride in Coptis by UPLC. The optimal conditions of separation and detection were achieved on an ACQUITY UPLC BEH-C18 column (2.1 mm×50 mm×1.7 μm) eluted with a gradient acetonitrile and 0.05 mol/L potassium dihydrogen phosphate solution at a flow rate of 0.2 mL/min, and detected at 350 nm. The results showed that there were good linear relationships (r=0.999 9) when the concentration of berberine hydrochloride and palmatine hydrochloride were in the range of 0.0372-0.2976 μg/mL and 0.0082-0.0656 μg/mL respectively, the recovery rates were more than 95%, and RSDs were less than 2%. This method was simple, rapid, accurate, and reproducible, and it can be used for determining the concentration of berberine hydrochloride and palmatine hydrochloride in Coptis.
